演題:KSHV-vGPCR-induced miR-34a expression promotes genomic instability by a broad suppression of genome maintenance mechanisms

抄録:

 Genomic instability is a critical, enabling hallmark of almost every cancer cell as it increases the naturally low cellular mutation rate greatly, for instance, by deregulating DNA replication, insufficient DNA repair or the failure to activate the DNA damage response. We show that the viral chemokine receptor vGPCR, encoded by the oncogenic Kaposi´s sarcoma-associated herpesvirus promotes genomic instability by broadly suppressing cellular mechanisms required for the maintenance of genome integrity including DNA replication, many DNA repair pathways and the DNA damage response. Their suppression is mediated by microRNA-34a that is induced by the vGPCR. Hence, our data support a protumorigenic, genomic instability-promoting role of miR-34a - so far predominantly seen as a tumor suppressor - and question the use of miR-34a mimetics for cancer therapy as they could dangerously affect genome integrity.
