エリザベート ガテフ博士は、1932年8月8日、ドイツ人とブルガリア人を両親としてブルガリア国ソフィア市に生まれた。1950年高等学校を卒業後、ブルガリアの敵国人と宣告され、高等教育から排斥され、1950年から1954年まで高電圧装置製造工場で金属加工の工員として働いた。1951年から1954年まで、働きながら機械工作専門の夜学に通い、エンジニアの資格を得た後、同工場で鍛造と金属加工機械の組み立てを行った。1956年2月には、西ドイツに移り住み、はじめは製図工として働いた。1956年11月から1962年11月の間に、ミュンヘン市のルードウィッヒ-マキシミリアン大学で生物学、化学および地理学を学び、高校教員資格を取得した。1962年12月には、結婚して米国オハイオ州クリーブランド市に移り住み、ここで、ケース−ウエスタン−リザーブ大学発生生物学センターの上級研究助手として、ハワード A. シュナイダ−マン教授の下で、昆虫発生のホルモン制御の研究に従事した。博士(Ph.D.)論文の研究(1968年から1972年)で、ショウジョウバエ(Drosophila melanogaster)の発生と遺伝の研究を行い、世界で最初のがん抑制遺伝子を発見した。1972年ドイツに戻り、フライブルグ大学でさらに12種類のショウジョウバエがん抑制遺伝子を発見した。博士は、この研究をもとに教授資格取得論文を完成し、フライブルグ大学の発生生物学の助教授(1981年)に、つづいて1982年、マインツ市のヨハネスグーテンベルグ大学の遺伝学の教授に任命された。マインツでは6種類のがん抑制遺伝子の分子クローニングを行い機能を解析した。すべてのショウジョウバエがん抑制遺伝子には、それに対応するヒトの相同遺伝子が存在している。ガテフ教授は、1988年癌研究マイエンブルグ賞(ハイデルベルグ市ドイツ癌研究センター)を、さらに1993年ドイツ癌学会よりドイツ癌賞を受賞された。

Outline of Personal History

Elisabeth Gateff was born in Sofia/Bulgaria on 8 August 1932 of german/bulgarian parentage. After finishing high school 1950, she was declared enemy of Bulgaria, and thus, was excluded from higher education. She worked from 1950 to 1954 in a factory for higher voltage electrical equipment as a metalcutter. Parallel to working, from 1951 to 1954, she attended evening classes specializing in machine construction. After obtaining an engineering degree, she constructed in the same factory forging and metalcutting instruments. February 1956, E. Gateff took residence in Western Germany where she initially worked as a draftswoman. From November 1956 to November 1962, she studies at the Ludwig-Maximilian-University, Münich. Biology, Chemistry and Geography, from where she obtained a high school teaching degree. December 1962 she took by marriage residence in Cleveland, Ohio, USA. Here she worked as Senior Research Assistant at the Developmental Biology Center, Case-Western Reserve University, Cleveland, Ohio, headed by Prof. Howard A. Schneiderman on the hormonal control of insect development. During her Ph.D.-thesis work (1968 to 1972), concerned with developmental and genetic studies of the fruit fly Drosophila melanogaster, she discovered the first tumor suppressor gene. Back in Germany (1972), at the University of Freiburg, twelve further Drosophila tumor suppressor genes were found. Based on this work she completed her Habilitation and was appointed, first Assistant Professor of Developmental Biology at the University of Freiburg (1981), and shortly thereafter (1982) Professor of Genetics, at the Johannes Gutenberg University in Mainz. In Mainz, six tumor suppressor genes were molecularly cloned and functionally analysed. All Drosophila tumor suppressor genes have corresponding human homologs. Prof. Gateff is the recipient of the 1988 Meyenburg Prize for Cancer Research (German Cancer Research Center, Heidelberg) and the 1993 German Cancer Prize given by the German Cancer Society.

エリザベートガテフ博士の初期の学問的興味は、植物群落生態学であった。ミュンヘン大学において植物学を専攻し、ババリア地方のアルプス山脈の河川、レック川中流域の植物群落の研究によって修士号を取得した。米国に移り住んだ後、オハイオ州クリーブランド市ケース-ウエスタン-リザーブ大学発生生物学センターのハワード A. シュナイダ−マン教授の研究グループに加わり、ここで、学問的興味は昆虫発生のホルモン制御の研究に移った。博士は、クリシュナ クマラン博士の指導の下、カイコガ(Hyalophora cecropia)発生のホルモン制御の研究を熱心に行い、エクダイソンの処理によって、表皮層やその他の器官におけるRNAや蛋白の発現レベルが劇的に増加することをオートラジオグラフィーの手法で示した。また、ホルモン精製過程でのそれぞれの精製標品中の幼若ホルモンの活性測定を行った。この頃、シュナイダ−マン教授の勧めで、三番目の研究分野であるショウジョウバエの発生の研究を始めた。博士は、当初、エルンスト ハドーン教授の考案による連続in vivo移植法を用いて、眼および触覚の成虫原基細胞の分化転換能力の研究を行った。この研究で、移植された眼の細胞は主として生殖細胞に分化転換する一方で、分化能を完全に失う場合もあることを見い出した。未分化の原基細胞は腫瘍化し致死的な増殖を示した。これらの観察は、lethal giant larvae (lgl) として知られる幼生致死性変異の発見へと発展した。ガテフ博士は、変異幼生の中に、脳の神経芽腫と成虫原基の腫瘍を発見した。これらの腫瘍は、ヒトの悪性腫瘍でみられるすべての特徴、すなわち、未分化で浸潤性をもち、原発部位でもあるいは野生型の成虫に移植後も自律的で致死性の増殖を示した。上述の腫瘍はホモ接合体の変異幼生にのみ発生することから、lgl 遺伝子はがん抑制遺伝子の定義に合致するものであった。このようにして、 最初のがん抑制遺伝子が同定され特徴づけられた。博士は、カリフォルニア大学アーバイン校で上記のショウジョウバエの結果を含むPh.D.論文を完成させ、ドイツに戻られた。フライブルグ大学では、ショウジョウバエゲノム上のその他のがん抑制遺伝子の可能性を検討し、徹底した突然変異導入実験から、12種のがん抑制遺伝子を発見した。これらの遺伝子の変異は、一種類か多くとも二種類の細胞系の悪性形質転換に関係しており、幼生の脳に神経芽種を起こす遺伝子もあれば、成虫原基の腫瘍の原因遺伝子もあり、さらに血球系の腫瘍や生殖細胞の腫瘍の原因遺伝子もある。これらすべての遺伝子は、それぞれの細胞の分化能に関わる発生制御遺伝子である。博士は、フライブルグで教授資格取得論文を完成して、マインツ市のヨハネスグーテンベルグ大学遺伝学主任教授に着任した後、ベルナルド メヒラー博士とともに6種類の遺伝子について構造と機能の解析を開始した。遺伝子のクローニングと構造解析、さらに遺伝子産物の解明から、細胞核や細胞質、細胞骨格系、ミトコンドリア、細胞間接着部位等の種々の細胞内諸器官における分布が明らかになった。予想される蛋白分子中の既知の機能ドメインから規定される細胞内分布は、6種類の遺伝子産物の機能について予備的な結論を出すのに役立った。たとえば、遺伝子のひとつは、恒常的にクロマチンの変化を及ぼして多くの遺伝子群を抑制的に制御するとして知られるポリコームグループ遺伝子群に属する。また、別の遺伝子はストレス蛋白ファミリーに属している。Lgl蛋白は大きい蛋白複合体の一部としてシグナル伝達機構で機能するとみられている。生殖細胞腫瘍を起こす遺伝子の遺伝子産物は、RNA結合能を保持するものと見られる。6種類の遺伝子はすべて少なくともマウスとヒトで相同遺伝子が存在している。このような事例と、遺伝子解析によって明らかにされた、多くのヒトやマウスの遺伝子がショウジョウバエのゲノム上にも存在するとする事実から、ショウジョウバエは、細胞のがん化の原因遺伝因子研究の最も良いモデルシステムである。

ガテフ教授は、研究と平行して多くの学生に遺伝学を講議された。また、多くの共同研究者の方々は、ショウジョウバエのがん抑制遺伝子とマウスおよびヒトにおけるその相同遺伝子の研究を続けておられる。博士は、1988年ハイデルベルグ市ドイツ癌センターの癌研究マイエンブルグ賞を受賞され、また、1993年にドイツ癌学会のドイツ癌賞を受賞された。

Academic Achievements

The early scientific interest of Prof. Elisabeth Gateff was plant sociology. At the University of Münich she specialized in botany, where she obtained her masters degree by studying the plant societies along the middle part of the alpine river Lech in Bavaria. After taking residence in the USA, she joined the group of Prof. Howard A. Schneiderman at the Developmental Biology Center, Case Western Reserve University, Cleveland, Ohio. Here her scientific interest shifted to a second field, namely the hormonal control of insect development. Under the supervision of Dr. Krishna Kumaran she participated in extensive investigations on the hormonal control of silk moth develpment. Autoradiographic studies demonstrated dramatically increased RNA and protein levels in the Cecropia epidermis and other organs when exposed to ecdysone. She also was involved in assayng the juvenile hormone activity of different hormone preparations during the purification of the hormone. She became initiated into a third field of research when Prof. Schneiderman suggested work on Drosophila development. Using the serial in vivo transplantation method, introduced by Prof. Ernst Hadorn, she studied initially the transdetermination capacity of the eye-antennal imaginal disc cells. These studies showed predominantly transdetermination of eye cells to genital cells, but also complete loss of the differentiation capacity of the serially transplanted cells. The undermined blastemas grew in a tumorous, lethal fashion. These later observations were extended by the discovery of the larval lethal mutation designated as lethal giant larvae (lgl). In the mutant larvae Prof. Gateff found a neuroblastoma growing in the brain and tumors of the imaginal discs. These tumors exhibited all characteristics of human malignant tumors, namely undifferentiated, invasive, autonomous lethal growth in situ as well as after transplantation into a wild-type adult host. Since the above tumors developed exclusively in the homozygous mutant larvae, the lgl gene met the definition of a tumor suppressor gene. Thus it represented the first identified and characterized tumor suppressor gene. Upon completing her PhD thesis at the University of California at Irvine, which included the above results with Drosophila, she returned to Germany. At the University of Freiburg she investigated the possible existence of additional tumor suppressor genes on the Drosophila genome. Extensive mutagenesis experiments yielded twelve independent tumor suppressor genes. Mutations in these genes are causally related to the malignant transformation of one or at the most of two specific cell-types. Some of the genes cause neuroblastomas in the larval brain, other genes imaginal disc tumors and still others blood cell tumors or gonial cell tumors. All genes represent developmental genes interfering with the differentiation capacity of the respective cells. After completing here habilitation in Freiburg and the appointment to the Chair of Genetics at the Johannes Gutenberg University in Mainz, together with Dr. Bernard Mechler, the molecular and function analysis of six of the genes was initiated. The cloning, the structural analysis of the genes, and their gene products revealed their presence in different cellular compartments, such as the nucleus, the cytoplasm, the cytoskeleton, the mitochondria and the cell-cell junction. The cellular localization added by the presence of well defined functional domains in the putative proteins allowed preliminary conclusions as to the possible function of the six proteins. One of the genes belongs to the Polycomb-Group genes which induce permanently chromatin changes, and thus, regulate negatively a great number of genes.
Another gene belongs to the family of stress proteins. The Lgl-protein is a part of a large protein complex which seems to function in signal transduction. The putative product of the gene inducing a gonail cell tumor seems to possess RNA-binding capacity. All six genes have at least two mouse and human homologs. This and the fact that many human and mouse genes are present also on the Drosophila genome, aided by powerful genetic methodology, makes Drosophila a prime model system to study the causal genetic factors in the malignant transformation of cells.

Parallel to her research activities Prof. Gateff has taught genetics to many student generations. A number of collaborators are continuing the work on the Drosophila tumor suppressor genes and their mouse and human homologs. She is the recipient of the 1988 Meyenburg Prize for Cancer Research given by the German Cancer Center in Heidelberg, and the 1993 German Cancer Prize presented by the German Cancer Society.
