略歴

嶋 昭紘博士は、1941年1月9日、京都市に生まれた。1965年3月に東京大学理学部生物学科(動物学課程)を卒業した。同年4月に東京大学大学院理学系研究科動物学専攻修士課程に進学し、秋田康一教授のもとでラットを使った放射線生物学の研究を開始した。1967年7月に東京大学助手(理学部動物学教室放射線生物学講座)となり、1971年1月に理学博士を授与され、秋田康一、江上信雄両教授のもとでメダカ化学発癌等の研究に従事した。1976年4月に滋賀医科大学助教授(放射線基礎医学教室)となり、DNA修復の研究を始め、1982年からは米国Brookhaven National LaboratoryのRichard B. Setlow博士の客員研究員を兼任し、自家樹立したキンギョ培養細胞と紫外線を使ったDNA修復の研究を発展させた。1985年12月から東京大学教授として理学部動物学教室放射線生物学講座を担当し、メダカを使った生殖細胞突然変異の研究を開始するとともに、日本メダカ野生集団の系統維持にあたってきた。動物学教室主任、東京大学総長補佐、日本放射線影響学会会長等を勤め、1996年12月から東京大学柏キャンパス新大学院研究科創設および推進プロデューサーとして先端生命科学領域の創設にあたり、1998年4月からは東京大学大学院新領域創成科学研究科教授として、メダカゲノムプロジェクトを組織し推進してきた。1999年からはメダカゲノム国際コンソシアムMedaka Genome Initiative (MGI) を主宰し、EMBOコース等でメダカ研究を国際化してきた。2003年3月に東京大学を退官し、現在は東京大学名誉教授である。

Outline of Personal History

Akihiro Shima was born on 9 January 1941 in Kyoto City, Japan. He graduated from the Faculty of Science, University of Tokyo in 1965. In 1967 he was appointed Research Associate in the Zoological Institute of the University of Tokyo, where he worked on radiation effects on the rat thymus, as well as on chemical carcinogenesis using the medaka, as a research collaborator with Prof. Yasukazu Akita and Prof. Nobuo Egami. In 1976, he left Tokyo for Shiga University of Medical Science as Associate Professor of experimental radiology. There, he started studying DNA repair using a cultured cell line he established from the goldfish. Since 1982, in a long collaboration as a Visiting Scientist with Dr. Richard B. Setlow of Brookhaven National Laboratory, Dr. Shima's investigations have been ongoing on DNA repair and mutagenesis using the medaka. In 1985, he returned to the University of Tokyo as Professor of radiation biology and launched his life work on medaka genome research; as a first step he established a specific-locus test system to study germ cell mutagenesis. After serving as Chairman of the Zoological Institute, Assistant to President of the University of Tokyo, and President of the Japan Radiation Research Society, he was appointed the founder of the new department in the new Kashiwa campus of the University of Tokyo. In 1998, he was appointed Founding Professor of Integrated Biosciences, Graduate School of Frontier Sciences of the University of Tokyo and has been serving as the PI of the Medaka Genome Project granted by the MEXT (Japan) as a Priority Area (#813). He has also represented an international medaka genome consortium, the Medaka Genome Initiative (MGI).
He had been married for 30 years before he lost his wife from sickness in 1999. He has one married daughter and one granddaughter.

研究業績

嶋 昭紘博士がメダカの化学発癌研究を初めて手がけたのは、1970年〜80年代に常陸宮殿下をはじめ、松島泰次郎博士、高山昭三博士、石川隆俊博士が、化学発癌研究におけるメダカの有用性を実証する諸成果を上げられていた時期であった。メダカの専門家である江上信雄教授の助手であった嶋博士は、江上研究室でメダカを用いた化学発癌実験を開始するのに先立ち、癌研究所へ石川隆俊博士を訪れ、ジエチルニトロソアミンの安全取り扱いを含めたメダカ化学発癌実験の基礎の教えを請うた。以後、江上研究室では、ジエチルニトロソアミンによるメダカ肝臓癌誘発過程での肝細胞の増殖動態研究が行われ、嶋博士はDNA顕微蛍光測光法を駆使した細胞動態解析を分担した。その後、常陸宮殿下と石川博士がアフリカ肺魚の自然発生腫瘍を記載された研究では、DNA顕微蛍光測光法によるゲノムサイズの測定を分担した。
嶋博士は、魚類の腫瘍からDNA修復へと研究を発展させ、特に紫外線によるDNA損傷とその修復を詳細に追究した。当時、コロニー形成しうる魚類培養細胞が存在しなかったので、嶋博士はキンギョ尾ひれの細胞を初代培養し、約2年間の継代培養により、コロニー形成可能なRBCF-1株を樹立した。この細胞を用いて、紫外線により細胞核DNAに形成されるピリミジンダイマー量と、細胞の生存率との間に直接的な定量関係があることを、魚類細胞に特有のDNA修復機構である光回復を用いて初めて証明した。嶋博士はこの成果により、DNA修復現象の発見者であるRichard B. Setlow博士のもとでBrookhaven National Laboratory客員研究員を兼務するに至り、現在の地球環境で注目されている紫外線UVBによるDNA損傷と修復に関する研究を行った。
嶋博士は1985年から、江上信雄教授の後任としてわが国メダカ野生集団の系統維持にあたり、メダカを新世代のモデル動物として確立する研究に着手した。その第一歩として、メダカ特定座位法を開発し、メダカの生殖細胞突然変異の研究を可能にした。メダカが卵生動物であり、しかも透明な卵膜を持つことを活かし、約480万の遺伝子座を調べて提唱した「総突然変異」は、マウス特定座位法では得られない高感度の遺伝指標である。さらに生殖細胞突然変異の分子機構の解析へと研究を進め、AP-PCR DNA フィンガープリント法をメダカゲノム解析に応用し、放射線による生殖細胞突然変異体には、ゲノムの広範囲にわたり欠失があることを実証した。また、減数分裂時のゲノムの変化をDNAフィンガープリント法で検出できることを発見し、ゲノム全般を網羅した遺伝子連鎖地図をメダカで初めて作製して、分子遺伝学・分子生物学に基づくメダカゲノム研究の端緒を開いた。
これまで、メダカの遺伝学研究はわが国の會田龍雄博士、山本時男博士、江上信雄博士、富田英夫博士ら諸先達により確立されてきたが、最近、嶋博士はメダカゲノム国際コンソシアム(MGI)を主宰してゲノム解析を進めている。これにより、メダカを新世代のモデル動物として確立する一方、メダカゲノム情報に基づいてメダカ化学発癌機構の解析が進展し、比較腫瘍学研究の重要な一翼を担うことが期待されている。

Academic Achievement

It was in the midst of successful demonstration of the usefulness of the medaka, Oryzias latipes, in carcinogenicity testing by Prince Masahito Hitachi, Drs. T.Matsushima, S.Takayama and T.Ishikawa in the 1970s and 80s that Dr. Akihiro Shima started his research on chemical carcinogenesis using medaka. Prior to launching the medaka carcinogenesis project, Professor N.Egami, who was the expert in medaka biology and Dr. Shima's supervisor, sent Dr. Shima to the Cancer Institute asking Dr. T.Ishikawa to instruct him in the basic protocols for carcinogenesis experimentation using diethylnitrosamine and medaka. Thereafter, a series of studies were performed in Prof. Egami's lab on cell population kinetics of medaka hepatocytes after treatment of medaka with the carcinogen, and Dr. Shima contributed to those studies through cell proliferation analyses using DNA microfluorometry. Later, Dr. Shima contributed also through microfluorometry to a study on a spontaneous neurinoma in an African lungfish by Prince Masahito Hitachi and Dr. Ishikawa.
Dr. Shima further stepped into studies of DNA repair using ultraviolet light and a cultured fish cell line capable of colony formation he established from the goldfish. The demonstration of the causal relationship between the amount of pyrimidine dimers formed by UV in nuclear DNA and cell survival rate using a photorepair system initiated a long-standing collaboration with Dr. R.B.Setlow. Their collaborations were focused on DNA damage by UVB and its photorepair, and on germ cell mutagenesis by heavy ion particles using the medaka.
On returning to the University of Tokyo as Professor of Radiation Biology in 1985, Dr. Shima was in charge of preserving wild populations of the Japanese medaka, and launched his life work on the study of the medaka genome. As a first step, he succeeded in developing the medaka specific-locus test system, which allowed him to propose the "total mutation," a genetic endpoint far more sensitive for germ cell mutagenesis compared with that available in mice. Application of AP-PCR DNA fingerprinting to detect genomic alterations in the progeny of paternally-irradiated medaka demonstrated genome-wide occurrence of DNA deletions. Furthermore, utilization of DNA fingerprinting for detection of genomic changes in backcross progeny between two inbred strains, one from the northern population and the other from the southern population of the Japanese medaka, successfully produced a first genome-wide genetic linkage map of the medaka. These results founded the firm basis for promoting medaka genomics based on molecular biology and genetics. In order to consolidate the basic medaka genetics developed by predecessors, Drs.T.Aida, T.Yamamoto, N.Egami and H.Tomita, efforts have been made on genome analysis in the framework of an international consortium, the Medaka Genome Initiative (MGI) led by Dr.Shima. Re-visiting usefulness of the medaka in carcinogenicity testing based on firm medaka genomics could be anticipated to promote studies on comparative oncology.
