要旨:Traditional IHC detects the presence of single epitopes with a resolution limit in the range of 200 nm (the diffraction limit of conventional light microscopy). As a result, it is only possible to describe the proteins in terms of expression level and co-localization, rather than interactions that occur on the order of tens of nanometers. The ability to interrogate for the presence and distribution of specific intermolecular interactions on tissue samples allows for IHC to transition from structural to functional diagnostics. The definition of a biomarker broadens to include protein interactions. Assays to measure these “signaling complexes” represent a missing component of the precision medicine toolbox. This presentation will describe a technology that allows us to view protein pairs, under a light microscope, to see if they are in close proximity and show in model systems that this test may be a predictor of drug response.  Multiplexed IHC applications will also be presented.
