演題:Formation of spindle and nuclear envelope on chromosomes – building completely different architectures using a similar principle

抄録:

Chromosome segregation and following nuclear formation are essential for cell proliferation. To support these events, chromosomes must organize the bipolar spindle during M phase and the nucleus during interphase. What is the principle behind organizing these completely different micron scale architectures on chromosomes? While the nucleosome, composed of ~146 bp DNA wrapping around octameric core histones, is the fundamental unit of chromosomes, and histones receive a variety of cell cycle-dependent post-translational modifications, it has been technically difficult to dissect direct roles of nucleosomes and histone modifications in these mitotic processes, since changes in transcriptions upon histone manipulations may affect these events indirectly. We have recently developed a method to reconstitute chromatin’s capacities to organize the spindle and the nuclear envelope in the absence of de novo transcription using recombinant histones and Xenopus egg extracts. Using this system, we established the direct roles of nucleosomes in these cell biological events. I will discuss the underlining principle that drives chromosome-induced assembly of the spindle and the nuclear envelop.
