演題:How the ubiquitin system manages genome stability upon DNA replication stress

Dealing with damage during DNA replication is important because the replication machinery cannot cope with defective templates. To avoid permanent arrest, cells employ mechanisms of damage bypass, allowing replication to “skip over” lesions without their actual removal. This ensures the completion of DNA replication but is often associated with damage-induced mutations. Therefore, DNA damage bypass is also a potential source of genome instability that can lead to cancer or ageing. In eukaryotic cells, the pathway is governed by the ubiquitin system, via mono- and polyubiquitylation of the essential replication factor PCNA. We have investigated the consequences of this modification in human cells and identified a surprising complexity of PCNA-based ubiquitin signalling events and synthetic lethalities with strong implications for the viability of BRCA1-deficient tumour cells.
