演題:Beyond oncogene addiction: In search of new therapeutic paradigms in cancer

抄録:
Oncogene addiction (i.e, the unusual dependence of cancer cells on the activity of an oncogenic protein) has served as an invaluable paradigm in oncology drug development. Consequently, most molecularly targeted agents that populate various drug development pipelines have been identified based on their ability to a) suppress the function of oncogenic drivers, and b) selectively kill cancer cells that express these activated oncoproteins. Importantly, the identification of novel driver oncogenes has relied extensively on both large-scale sequencing efforts that have helped to uncover functionally significant variants, as well as genetic dropout screens. As relevant data generated from these projects reach saturation, the utility of the oncogene addiction paradigm faces its own saturation risks. Therefore, there is an increasing need to explore alternative concepts for the identification of new targets. Synthetic lethality and lineage addiction are additional therapeutic paradigms that have demonstrated varying degrees of success in identifying clinically viable targets. My group has been interested in exploring two new concepts to uncover cancer-selective vulnerabilities: oncogene overdose, and targeted oncoprotein mislocalisation. I will discuss our progress in these two areas focusing on how glucose availability can influence oncogenic output, and how to use trafficking perturbants to drive cancer-selective cell killing.
