抄録:The mammalian nucleus forms subnuclear compartments marked by diverse subnuclear structures such as nuclear bodies and histone modifications. These subnuclear compartments are intrinsically associated with gene regulation and 3D genome organization. To date, our ability to directly map and understand organization of single nuclei in tissues has been limited due to the lack of high-resolution spatial multi-omics technology. Here, we introduce a new imaging-based technology (two-layer DNA seqFISH+), enabling simultaneous mapping of 100,049 genomic loci, together with nascent transcriptome and various subnuclear structures in single cells in the adult mouse cerebellum. We identified cell-type specific subnuclear compartments, which can shape structural hubs for radial chromosomal positioning and inter-chromosomal interactions in neurons and glial cells. Together, our results provide a major step forward in understanding single-cell nuclear architecture within complex tissues.
